RESONANCE RAMAN SPECTROSCOPY

The Resonance Raman Spectroscopy facility is part of the Bioenergetics, Structural Biology and Mechanisms unit (UMR 8221 / CNRS & iBiTec-S / CEA) at CEA Saclay. It provides users with advanced Raman spectrometry and responds to most of the needs of vibrational analyses.

The Raman platform includes 7 spectrometers with many accessories. Broad temperature range possible (thermostats 273-320 K; cryostats 4-250 K). Samples in a wide range of physical forms can be analysed, in particular those containing pigmented molecules (see below). The laboratory is specialised in particular in resonance Raman spectroscopy of pigment cofactors in biology(carotenoids, chlorophylls, hemes, flavins …), including expertise in in vivo investigations of biochemical, regulatory and photo-induced reactions.

Equipment:

-2 Yobin-Jvon U1000 spectrometers; double monochromator; highly-sensitive, liquid-N2-cooled CCD detector. One optimised in the red and the other in the blue region of the spectrum. These are coupled to a number of lasers giving excitations throughout the visible, near-UV and near-IR range.

-2 other Yobin-Jvon U1000 double monochromator spectrometers (N2-cooled CCDs), coupled to confocal microscopes for Raman imaging measurements. Excitation throughout the visible.

-1 Yobin-Jvon T64000 spectrometer (single monochromator; N2-cooled CCD), excitation throughout the visible.

-2 Bruker FT-Raman spectrometers (Bruker IFS66 FTIR + Bruker FRA106 Raman module; N2-cooled Ge diode detector), excitation 1064 nm (also possible at other near-IR wavelengths).

Sample state (liquid, powder, gel, solid …) is only limited by signal size (presence of pigmented molecules required for studies in complex media). Recent measurements include studies of regulatory processes in photosynthetic membranes in vivo (whole leaves and whole microorganisms), and molecular structure of carotenoids and opsins in ex vivo human retina.
