This course will provide the participants with an overview of strategies for preparation and characterization of macromolecular complexes in view of biochemical and structural studies. Topics will cover top down (purification from cells expressing proteins under the control of their natural regulatory elements) and bottom up (reconstitution from recombinant subunits assembled in vitro or co-expressed) approaches for sample preparation as well as various approaches for the characterization of macromolecular assemblies (composition, homogeneity/stability and architecture). The course is intended primarily for post-doctoral researchers, PhD students and researchers in structural biology. The goal is that the participants improve their theoretical background and gain experience in designing experimental strategies relevant for their projects.
