Cryo Electron Microscopy (Cryo-EM)

The EM platform is composed of:
An electron microscope JEOL 1400 equipped with a Lab6 operated up to 120 keV.
An electron microscope Glacios (Thermofisher) operated at 200 kV
An EM-GP2 plunger (Leica) to freeze cryo-grids.
A cryogenic FIB-SEM (Zeiss), combining a focused galium ion beam with a scanning electron microscope Gemini 2

The EM platform proposes different services:
Negative stain:
–        Characterize a sample (proteins/bacteria/viruses)

–        Evaluate the quality of the sample for further cryo-EM analysis
 –        Allow a 15-20A resolution 3D reconstruction of protein/protein complexes.
Cellular electron microscopy:
–        Observation of sections of embedded samples (bacteria, cells)

 –        Observation of fixed bacteria, phages, virus
Cryo-EM imaging:
–        Evaluation of the cryo-EM grid (Ice thickness, Protein distribution)
–        First images for 2D classification and low resolution 3D map
Cryo-FIB-SEM
Capture a snapshot of cellular structures at near native states (also named high resolution volume EM)
