HIGH THROUGHPUT SCREENING

The high throughput crystallization platform offers screening and optimization techniques, including vapor diffusion and crystallization under oil, in sitting-drop format. Flexible methods have been developed to adapt each step to individual project requirements.

Application: Crystallization is performed using commercially available screening kits, as well as customized screens prepared from in-house stock solutions. A wide variety of robotic protocols are available to optimize crystal growth and diffraction quality.

Equipments:  Freedom Evo Workstation (Tecan), Mosquito Crystal (SPT Labtech).

Description: Homemade and optimization screens are prepared in a 96-well format with a Freedom Evo Workstation, sitting drops -from 100nl to 500nl- are dispensed using a Mosquito Crystal robot.

Protocols for automated micro-seeding and variations in droplet ratios are used routinely.

AUTOMATED STORAGE AND VISUALIZATION

Plates are stored and automatically imaged at 20°C in an automated imaging system equipped with UV light. Images are saved on a database accessible to users via a web-interface.

Application: images of crystallization drops are captured at different time points to follow the nucleation and growth of protein crystals. UV fluorescence can help distinguish protein crystals from salt crystals or can help reveal tiny crystals that are difficult to identify in visible-light images.

Equipments: Rock Imager 360 UV (Formulatrix).

Description: the Rock Imager 360 UV canmonitor crystallization experiments over time, capturing high-quality images of crystallization drops under visible and UV light. The Rock Maker software is a complete Laboratory Information Management System (LIMS) equipped with an AI-based autoscoring system.
